Skin is the third most zinc (Zn)-abundant tissue in body, In skin, mast cell (MC) granules are enriched Zn. Cellular Zn levels strictly regulated by two groups of transporters: transporters (ZnTs) and Zrt-, Irt-like protein (ZIPs). ZnT expressions were previously determined bone-marrow derived MCs (BMMCs). contrast, little known about ZIP expressions. Thus, aim this study to determine murine MCs. Rodent categorized into connective (CTMCs) mucosal (MMCs). The former latter presumed correspond human tryptase- chymase-positive (MCTC) tryptase-positive (MCT), respectively. CTMCs MCTC constitutively distributed gut lamina propria, pulmonary perivascular tissues, whereas MMCs MCT epithelium lungs. Fetal skin-derived (FSMCs) which obtained as follows; skin from fetal mice was enzymatically digested with trypsin prepare single-cell suspensions. Cells cultured a medium containing IL-3 stem factor for three weeks. Non-adherent cells collected purified using c-kit microbeads.
